What Independent Lab Testing Actually Checks on a Peptide Batch

Reference

Reference Glossary

Plain, research framed definitions of the terms a careful buyer meets on a certificate of analysis and around the laboratory bench.

Basics

3
Peptide
A short chain of amino acids joined in a defined sequence, shorter than a full protein. In a research setting each peptide is identified by its sequence and studied as a discrete compound.
Amino acid
One of the small building block molecules that link together to form peptides and proteins. The order in which they are arranged gives a peptide its identity.
mg versus mcg
Milligrams and micrograms are units of mass, with one milligram equal to one thousand micrograms. Keeping the two straight avoids thousandfold errors when calculating concentration and volume.

Chemistry

3
Peptide bond
The chemical link that joins one amino acid to the next along a peptide chain. The number and arrangement of these bonds define the length and structure of the molecule.
Concentration (mg/mL)
The amount of peptide present in each millilitre of solution, found by dividing the peptide mass by the solvent volume. It is the reference figure for working out how much solution corresponds to a given quantity.
Molecular weight
The mass of a single peptide molecule, usually reported in daltons and derived from its amino acid sequence. It is used to relate mass to the number of molecules present in a sample.

Handling

4
Lyophilised powder
A peptide that has been freeze dried into a stable dry solid for storage and transport. It is supplied in this form and brought into solution at the bench before measurement.
Reconstitution
The laboratory step of dissolving a lyophilised powder in a suitable solvent to produce a solution of known concentration. Careful reconstitution is the basis for any accurate volume measurement.
Bacteriostatic water
Sterile water containing a small amount of preservative that limits microbial growth, commonly used as the solvent for reconstituting research peptides. The preservative helps a reconstituted sample stay stable while it is stored between measurements at the bench.
Storage and cold chain
The set of temperature controlled conditions that keep a peptide stable from manufacture through to the bench. Maintaining the cold chain protects the integrity of both the dry powder and any reconstituted solution.

Testing

6
Certificate of analysis
A document supplied with a batch that records the analytical results confirming a compound's identity and measured purity. It is the primary paperwork a careful research buyer reviews before accepting a sample.
HPLC purity
A purity figure produced by high performance liquid chromatography, where the sample is passed through a column that resolves it into distinct peaks and the target compound is reported as a percentage of the total signal detected. A higher figure means fewer other peaks were picked up in that run.
Endotoxin screening
A laboratory test that checks a sample for bacterial endotoxins, contaminants that can appear during manufacturing. It is one of the identity and quality checks recorded on supporting documentation.
Batch and lot number
A unique identifier assigned to a specific production run so that a sample can be traced back to its analysis. Matching this number to its paperwork is how a batch is confirmed.
Public verification register
An openly accessible record where a batch identifier can be checked against its published analysis. It lets a research buyer confirm documentation independently of the supplier's own claims.
Identity confirmation
The step of establishing that a supplied compound matches its stated amino acid sequence, typically through analytical methods such as mass spectrometry. It sits alongside purity as a core documentation check for a research buyer.
